Background 4-1BB Ligand/TNFSF9 is expressed on activated B cells, macrophages, and dendritic cells. 4-1BB Ligand-expressing cell lines or soluble 4-1BB Ligand induce mitogen-activated, CD3-cross-linked, or alloreactive T cells to proliferate and secrete IL-2 in vitro. 4-1BB Ligand is important for the generation of antiviral CD8 T cell responses[1]. 4-1BB Ligand enhances proliferation, cytokine production, and CTL effector function of TCR transgenic T cells. Both CD4 and CD8 T cells respond to 4-1BB ligand (4-1BBL) with similar efficacy[2]. The interaction of 4-1BBL with 4-1BB provides costimulatory signals to both CD4+ and CD8+ T cells through the activation of NF-κB, c-Jun and p38 downstream pathways. 4-1BBL can enhance antitumor responses in the absence of CD28, however for protective immunity, CD28:B7 signaling is necessary[3]. |
