Cytosolic Apaf-1 f o r ms an oligomeric complex with caspase-9 in the presence of cytochrome c a n d dATP, which leads to caspase-9 activation a n d subsequent activation of caspase-3 (2, 3). Oligomeric Apaf-1 a n d pro-caspase-9 bind to each other via their respective NH2-terminal CARD domains, consequently enhancing the catalytic activity of caspase-9 (4), leading to activation of the caspase signaling cascade a n d apoptosis.
